**  Cirm Northern Blots  **

Introduction

Northern Blots from CirmBlots'  Human Multiple Tissue Northern Blots (HMTN) contains RNA from 50 hard-to-obtain human, rats, mouse adult and fetal tissues and is carefully normalized to the mRNA expression levels of eight housekeeping genes for accurate assessment of target genes expression.  It allows researchers to quickly determine the tissue distribution, size, and splice forms of particular gene transcripts in a number of hard-to-obtain tissues.

Both pre-made total RNA and poly A⁺ RNA Blots (mRNA) are available right now from CirmBlots, CirmBlots Northern Blots.  The total RNA blots are used for quick screening of the tissue distribution of your genes, for examining the tissue distribution of a highly expressed gene, or for determining the tissue distribution of an Expressed Sequence Tag in order to know from which library to clone the gene.  The CirmBlots' poly A⁺ RNA is purified by several passages through oligo (dT) cellulose columns, runs on a denaturing formaldehyde per 1.2% agarose gel, and blotted onto a positively charged nylon membrane.  Each lane contains approximately 2-3 Micro-gram of pure Poly A⁺ RNA from specific tissues.  The Poly A⁺ Northern Blots are used to obtain A Picture Perfect@ Northern for publication purposes.  These blots are also used to examine the tissue distribution of a low abundance gene and to study multiple forms of mRNA due to alternative splicing.

The CirmBlots RNA loaded in each lane is adjusted so that a visible ß-actin signal is present in every lane.  The nylon membrane allows multiple re-probing to be performed when the blot is handled and stripped properly.  We have been able to successfully strip and re-probe the Blots several times.  For size estimation, ink marks indicate the location of the RNA size markers.  Blots are most suitable for detecting transcripts in the range of 0.5-10 kb; transcripts outside this range may not be present.  A complete User Instruction/Manual, human ß-actin cDNA probe, and trial-size (25-ml) Express Hyb; Hybridization Solution is included with all blots.  The human ß-actin probe cross hybridizes with mouse and rat.
